A Fine-Structure Map of Spontaneous Mitotic Crossovers in the Yeast Saccharomyces cerevisiae

چکیده
Homologous recombination is an important mechanism for the repair of DNA damage in mitotically dividing cells. Mitotic crossovers between homologues with heterozygous alleles can produce two homozygous daughter cells (loss of heterozygosity), whereas crossovers between repeated genes on non-homologous chromosomes can result in translocations. Using a genetic system that allows selection of daughter cells that contain the reciprocal products of mitotic crossing over, we mapped crossovers and gene conversion events at a resolution of about 4 kb in a 120-kb region of chromosome V of Saccharomyces cerevisiae. The gene conversion tracts associated with mitotic crossovers are much longer (averaging about 12 kb) than the conversion tracts associated with meiotic recombination and are non-randomly distributed along the chromosome. In addition, about 40% of the conversion events have patterns of marker segregation that are most simply explained as reflecting the repair of a chromosome that was broken in G1 of the cell cycle.

Initiation of Ageing Process by Meiotic and Mitotic Recombination within the Ribosomal DNA Genes in Saccharomyces cerevisiae
In the budding yeast of Saccharomyces cerevisiae the tandem repeated of rDNA genes are located onchromosome XII, which is in the nucleolus. There are different types of proteins in the nucleoluskeleton, silencing proteins have got important role in nucleolus. It is shown that meiotic recombination between nonsister chromatids in the rDNA genes are stronglysuppressed, and s...
متن کاملSaccharomyces Cerevisiae as a Biocatalyst for Different Carbonyl Group under Green Condition
In this researchsaccharomyces cerevisiae (baker’s yeast) was used as a cheap, readily accessible, selective, efficient, and green bio-catalyst in a chemo selective reduction of carbonyl group to hydroxyl group. In this green procedure three substrates e.g. (3-(3-nitrophenyl)aziridin-2-yl)-1-phenyl-methanone, pyruvate ester, and 2-acetyl-γ-butyrolactone were r...
متن کاملFrom the Cover: mitotic gene conversion events induced in G1-synchronized yeast cells by gamma rays are similar to spontaneous conversion events.
In a previous study, we mapped spontaneous mitotic reciprocal crossovers (RCOs) in a 120-kb interval of chromosome V of Saccharomyces cerevisiae. About three-quarters of the crossovers were associated with gene conversion tracts. About 40% of these conversion tracts had the pattern expected as a consequence of repair of a double-stranded DNA break (DSB) of an unreplicated chromosome. We test th...
متن کاملA Simple and Rapid Protocol for Producing Yeast Extract from Saccharomyces cerevisiae Suitable for Preparing Bacterial Culture Media
Yeasts, especially Saccharomyces cerevisiae, are one of the oldest organisms with broad spectrum of applications, owing to their unique genetics and physiology. Yeast extract, i.e. the product of yeast cells, is extensively used as nutritional resource in bacterial culture media. The aim of this study was to develop a simple, rapid and cost benefit process to produce the yeast extract. In this ...
